Cloning and expression analysis of a novel zinc transporter gene from Nasturtium officinale

ZIP (Zinc-regulated transporter, Iron-regulated transporter-like Protein, ZIP) plays an important role in uptake or transport Zn Fe plants. So studying ZIPs plant can help to elucidate the molecular mechanism for and transportation. Herein, we have cloned a novel transporter gene NoZIP3 from Nasturtium officinale root using rapid-amplification of cDNA ends (RACE) method, evaluated its function by functional complementation assay determined relative transcriptional level quantitative real-time Reverse Transcription Polymerase Chain Reaction (qRT-PCR). The results showed that full-length was 1029 bp, which encoded polypeptide 342 amino acids with conserved domain typical ZIPs. sequence shared high similarity (57–89%) other species. could complement mutant yeast deficient transportation; however, it not qRT-PCR revealed roots much higher than leaves under deficiency, deficiency sufficient nutrition. These suggested might be preferentially expressed had roles transportation, provide foundation further characteristics this gene.
